Platelet-rich plasma is a concentrate of platelets, which can be obtained from the patient’s whole blood during a proper procedure of centrifugation and isolation of this fraction. PRP is an autologous concentrate of platelets suspended in a small amount of plasma.
Platelet-rich plasma is also called a concentrate of growth factors (plasma rich growth factors – PRGFs), due to a high concentration of growth cytokines.
After the obtained PRP has been injected into tissue, the process of α – granules degranulation in platelets begins. Growth factors are released. The majority of growth factors are released by the platelets within the first hour.
Growth factors have a tremendous regenerative potential. They are responsible for the increase of collagen synthesis, cell mitosis and the growth of blood vessels. They stimulate the migration of other cells to damaged places in tissue and they induce the process of cell differentiation into specialised cells that have specific functions in the tissue.
After coming through the stem cell membrane, the growth factors immediately connect to the nearest cell membranes in the receptor region. In turn, transmembrane receptors activate the intracellular signalling pathway, which influences the expression of proper genes.
After a quick release of growth factors, the platelets contained in PRP synthesise and emit their additional amount for the next 7 days. After that time, the thrombocyte dies and the healing stimulation functions are taken over by macrophages..
The important fact is that the growth factors do not penetrate the cells, and therefore there is no risk of mutagenesis. We can only talk here about the stimulation of a regular healing process.
